INTENDED USE
DiaRT One Step RT - PCR kit is applied in public health and clinical laboratories for the qualtitative determination of virus RNA in health samples. The user has to provide for the specific primers and fluorescence marked probes that necessary to the detection of the genetic segment which is tipical of the virus is wanted to be determined. The DiaRT One Step RT - PCR kit contains all the required component for the construction of the successful reverse transcription and real-time PCR reaction. The user has to take into account the directives of own specific primers and fluorescent marked probes before optimize the reaction. This process is conduced by MgCl2 solution - the required amount can be measure into the reaction mix - which is part of the kit.
TEST PRINCIPLE
The reverse transcription means the process, which results in DNA replica by right of RNA template by the help of reverse transcriptase enzyme. This process in nature is tipical of Retroviruses. The RT-PCR - reverse transcription polymerase chain reaction - is an enzyme mediated process, which is adequate for specific multiplying ribonucleic acids like viral (chromosome, genofor) genetic code. The first step is in-vitro reverse transcription and the second step is periodic replication. Result of reverse transcription is single strand cDNA and the cDNA is multiplying during cycles. Both steps of PCR reaction happen at determined temperature. The first step of PCR is the starting denaturation of DNA, the second step is the amplification which is regulated by primers. During every cycle of amplification happen the denaturation-annealing-elongation process. The product of PCR cycles is the multiplyed DNA. The PCR reaction is regulated by users specific primer-probe oligonucleotid pairs.
